PROMOTILITY:

Description: Domperidone is an investigational antiemetic to be used with cancer chemotherapy. It has also shown efficacy in the treatment of gastroesophageal reflux disease (GERD).

Actions: antagonist at dopaminergic receptors, similar to phenothiazines and metoclopramide.

Uses: under investigation for treatment of chemotherapy-induced nausea/vomiting; potentially useful in gastroesophageal reflux disease (GERD).

Distinguishing Features: Domperidone crosses the blood-brain barrier more poorly than phenothiazines or metoclopramide, thus, extrapyramidal reactions are uncommon. Unlike metoclopramide, effects of domperidone on gastric motility are not antagonized by atropine. Bioavailability is 15%; T« ranges 7-8 hours.

Major Adverse Reactions: hyperprolactinemia;

Usual Adult Dosage: doses of 40-120 mg PO have been used in the treatment of gastric hypomotility.
