methdilazine / DilosynTM

ANTIPSYCHOTIC

Description: Methdilazine is an oral HA-blocker. It is a member of the phenothiazine HA-blocker group. Methdilazine is used in the treatment of allergic conjunctivitis and rhinitis, angioedema, pruritus, and urticaria. HA-blockers competitively antagonize histamine at the HA-receptor and antagonize the pharmacologic effects of histamine. They also have anticholinergic/drying activity, antipruritic effects, and sedative effects. Methdilazine has a high antihistaminic effect, high anticholinergic effect, low sedative effect, and very high antiemetic effect. This drug was approved by the FDA in 1960.

Contraindications: Infants, nursing mothers, narrow-angle glaucoma, stenosing peptic ulcer, prostatic hypertrophy, asthmatic attacks, bladder neck obstruction, pyloroduodenal obstruction, MAO inhibitors, cardiovascular disease, liver dysfunction.

Drug Interactions: MAO inhibitors: MAOIs can prolong and intensify the anticholinergic effect of HA-blockers. Ethanol, CNS depressants: Additive CNS depression.

Major Adverse Effects: Drowsiness, epigastric distress, thickening of bronchial secretions, postural hypotension, palpitations, sedation, dizziness, fatigue, lassitude, confusion, anorexia, urinary frequency, dysuria, photosensitivity.

Dosage: For treatment of allergic rhinitis, angioedema, urticaria, and pruritus associated with nonallergic conditions: Regular tablets and syrup Adults: 8 mg PO q6-12 hours as needed.

Children >3 years old: 4 mg PO bid-qid as needed

Chewable tablets Adults: 7.2 mg PO q6-12 hours as needed.

Children >3 years old: 3.6 mg PO bid-qid as needed.
